目的:采用超高效液相色谱(UPLC)建立了同时测定养血清脑颗粒中绿原酸、咖啡酸、阿魏酸和迷迭香酸4个酚酸类化合物的分析方法。方法:采用Waters Acquity UPLC系统,使用Waters HSS T3(100 mm×2.1 mm,1.8μm)色谱柱,流动相为0.01%甲酸水溶液-乙腈,梯度洗脱,流速0.4 mL·min-1,检测波长325 nm。结果:4个酚酸类化合物色谱响应线性范围良好(r≥0.9994);平均回收率(n=6)为97.6%~105%,RSD为1.2%~4.6%。结论:使用UPLC作为养血清脑颗粒中4个酚酸类成分的含量测定方法,可以作为养血清脑颗粒质控方法。且不同批次养血清脑颗粒中4个酚酸含量基本一致。
OBJECTIVE: To establish a method for the simultaneous determination of 4 phenolic acids in caffeine, caffeic acid, ferulic acid and rosmarinic acid in Yangxue Qingnao Granules by ultra performance liquid chromatography (UPLC). Methods: The Waters Acquity UPLC system was used. The mobile phase consisted of 0.01% formic acid in water with gradient elution at a flow rate of 0.4 mL · min-1 using a Waters HSS T3 (100 mm × 2.1 mm, 1.8 μm) nm. Results: The linear range of the four phenolic compounds was good (r≥0.9994). The average recoveries (n = 6) were 97.6% -105% with RSDs of 1.2% -4.6%. Conclusion: UPLC as a method for the determination of 4 phenolic acids in Yangxue Qingnao Granules can be used as a quality control method for Yangxue Qingnao Granules. And different batches Yangxue Qingnaogu particles in the four phenolic acids content is basically the same.
