目的观察紫苏油对大鼠慢性脑低灌注损伤所致认知损害的保护作用。方法将50只♂SD大鼠经双侧颈动脉结扎建立慢性脑低灌注损伤模型,随机分为假手术组、模型组和紫苏油高、中、低剂量组;紫苏油高、中、低剂量组分别ig 0.8、0.4、0.2mL.(100 g)-1紫苏油60 d后,进行Morris水迷宫实验,检测其空间记忆能力;然后,取大鼠海马及皮层组织测定其乙酰胆碱酯酶(TChE)的活力及乙酰胆碱转移酶(ChAT)的含量。结果 0.8、0.4、0.2 mL.100 g-1紫苏油均可改善慢性脑低灌注损伤大鼠学习记忆障碍,大鼠的TChE活性降低,ChAT活性升高(P<0.01或P<0.05)。结论紫苏油能明显减轻大鼠慢性脑缺血所致的认知功能障碍,抑制海马及皮层TChE的活性、增加ChAT的活性。
Objective To observe the protective effect of perilla oil on cognitive impairment induced by chronic cerebral hypoperfusion in rats. Methods Fifty male SD rats were ligated with bilateral carotid arteries to establish a model of chronic cerebral hypoperfusion injury. They were randomly divided into sham operation group, model group and perilla oil high, medium and low dose groups. Perilla oil high, Low dose group were ig 0.8,0.4,0.2 mL. (100 g) -1 perilla oil for 60 days, the Morris water maze test to detect the spatial memory capacity; and then take the rat hippocampus and cortical tissue measured acetylcholine Enzyme (TChE) activity and acetylcholine transferase (ChAT) content. Results 0.8, 0.4 and 0.2 mL of 100 g-1 perilla oil all improved learning and memory deficits in rats with chronic cerebral hypoperfusion injury. The activity of TChE and the activity of ChAT in rats were decreased (P <0.01 or P <0.05). Conclusion Perilla oil can significantly reduce the cognitive dysfunction induced by chronic cerebral ischemia in rats, inhibit the activity of TChE and increase the activity of ChAT in hippocampus and cortex.
