目的探讨氟伐他汀对高糖诱导的人足细胞nephrin和血管紧张素Ⅱ(AngⅡ)表达的影响。方法体外培养人足细胞,随机分为正常糖对照组(A组)、DMSO组(B组)、高糖组(C组)、氟伐他汀10-7、10-6、10-5 mmol/L处理组(分别为D1、D2、D3组)及高糖+氟伐他汀组(E组)。用Westernblot和放射免疫法分别检测足细胞nephrin和AngⅡ的表达水平。结果 B组、D1组、D2组nephrin表达与A组比较差异无统计学意义(P>0.05),而D3组nephrin表达较A组明显减少(P<0.05)。与A组相比,C组人足细胞AngⅡ表达增加,nephrin表达减少,并呈时间依赖性(P<0.05);而E组能明显逆转C组上述指标的变化(P<0.05)。结论氟伐他汀能抑制高糖环境下足细胞AngⅡ表达,上调nephrin表达,起到保护足细胞的作用。
Objective To investigate the effect of fluvastatin on the expression of nephrin and angiotensin Ⅱ (AngⅡ) induced by high glucose in human podocytes. Methods Human podocytes were cultured in vitro and randomly divided into normal control group (A group), DMSO group (B group), high glucose group (C group), fluvastatin 10-7,10-6,10-5 mmol / L treatment group (D1, D2, D3 group) and high glucose + fluvastatin group (E group). Western blot and radioimmunoassay were used to detect the expression of nephrin and AngⅡin podocytes respectively. Results There was no significant difference in nephrin expression between group B, group D1 and group D2 (P> 0.05), but nephrin expression in group D3 was significantly lower than that of group A (P <0.05). Compared with group A, the expression of AngⅡ and the expression of nephrin in group C were decreased in a time-dependent manner (P <0.05), while group E could reverse the change of the above parameters in group C (P <0.05). Conclusion Fluvastatin can inhibit Ang Ⅱ expression in podocytes under high glucose and up-regulate the expression of nephrin, which plays a role in protecting podocytes.
