目的探讨外源性瘦素对人肝癌细胞HepG2生长增殖的影响。方法体外培养HepG2,给予不同浓度瘦素(50ng/ml、100ng/ml、200ng/ml),MTT法观察瘦素对HepG2生长的影响,流式细胞仪检测瘦素对HepG2细胞周期的作用。结果 0ng/ml~200ng/ml瘦素在作用72h内呈现出剂量、时间依赖性地促进HepG2的增殖,不同浓度组之间及不同时间组之间,差异均具有统计学意义(P<0.01,P<0.01),并且随着瘦素浓度的增加,HepG2G0~G1期细胞比例逐渐下降(P<0.01),S期及G2-M期细胞比例逐渐上升(P<0.01,P<0.01)。结论瘦素在离体实验中具有明显的促进入肝癌细胞HepG2增殖的效应,此效应与促进细胞DNA合成、增强有丝分裂有关,提示瘦素有可能参与肝癌的发生、发展过程。
Objective To investigate the effect of exogenous leptin on the growth and proliferation of human hepatoma HepG2 cells. Methods HepG2 cells were cultured in vitro. Leptin (50ng / ml, 100ng / ml, 200ng / ml) was given in different concentrations. The effect of leptin on the growth of HepG2 cells was observed by MTT assay. The effect of leptin on the cell cycle of HepG2 cells was detected by flow cytometry. Results Leptin of 0ng / ml ~ 200ng / ml showed a dose - dependent manner for 72h, which could promote the proliferation of HepG2 in a time - dependent manner. The difference was statistically significant between different concentration groups and different time groups (P <0.01, (P <0.01). The percentage of cells in the phase of 0 ~ G1 phase in HepG2G decreased gradually with the increase of leptin concentration (P <0.01), and the percentage of cells in S phase and G2-M phase increased gradually (P <0.01, P <0.01). Conclusion Leptin has an obvious effect on promoting the proliferation of HepG2 hepatoma cells in vitro. This effect is related to the promotion of DNA synthesis and mitosis, suggesting that leptin may be involved in the occurrence and development of hepatocellular carcinoma.
