目的:探讨β-榄香烯治疗胃癌的作用机制。方法:应用四唑蓝(MTT)比色法、端粒重复扩增(TRAP)-微孔板杂交法、光镜检查和DNA末端原位标记染色法(TUNEL)研究β-榄香烯对胃癌细胞株SGC-7901(中分化)、MKN-45(低分化)和MKN-28(高分化)的细胞毒作用及其对端粒酶活性、细胞形态学变化和细胞凋亡的影响。结果:β-榄香烯对不同分化程度的胃癌细胞均具有较强的细胞毒作用;其抗癌作用与抑制端粒酶活性和诱导凋亡有关。β-榄香烯抑制端粒酶活性及诱导凋亡的效果与作用时间、浓度及细胞分化程度相关。结论:β-榄香烯对胃癌细胞具有很强的细胞毒作用,这一作用与抑制端粒酶活性和诱导凋亡有关。
Objective: To explore the mechanism of β-elemene in the treatment of gastric cancer. METHODS: Using tetrazolium blue (MTT) colorimetry, telomere repeat amplification (TRAP)-microplate hybridization, light microscopy, and TUNEL to study β-elemene on gastric cancer The cytotoxicity of cell lines SGC-7901 (medium differentiation), MKN-45 (low differentiation), and MKN-28 (highly differentiated) and their effects on telomerase activity, cell morphology, and cell apoptosis. RESULTS: β-elemene had a strong cytotoxic effect on gastric cancer cells with different degrees of differentiation. Its anticancer effect was related to inhibition of telomerase activity and induction of apoptosis. The effect of β-elemene in inhibiting telomerase activity and inducing apoptosis is related to the action time, concentration and cell differentiation. CONCLUSION: β-elemene has a strong cytotoxic effect on gastric cancer cells, and this effect is related to the inhibition of telomerase activity and induction of apoptosis.
