目的探讨表柔比星抑制乳腺癌细胞中c-FLIP表达的时间窗和量效关系。方法将MCF-7及MDA-MB-231乳腺癌细胞株分成两组:处理组中加入终浓度分别为4.0,2.0,1.0,0.5,0.25 mg/L的表柔比星,对照组加入等量生理盐水。各组分别作用24,48,72 h后采用RT-PCR技术检测两组细胞株中c-FLIP的表达,流式细胞仪检测细胞凋亡百分率。结果随着表柔比星浓度的增加和作用时间的延长,c-FLIP的表达整体呈下降趋势;随着表柔比星浓度的升高及作用时间的延长,乳腺癌细胞凋亡率逐渐增加,呈剂量和时间依赖性。其中以浓度为2 mg/L作用72 h凋亡率最高。结论表柔比星化疗诱导乳腺癌细胞的凋亡可能系通过抑制c-FLIP的表达,且其效果以浓度为2 mg/L作用72 h时为最佳。
Objective To investigate the time-window and dose-response relationship of epirubicin in inhibiting c-FLIP expression in breast cancer cells. Methods MCF-7 and MDA-MB-231 breast cancer cell lines were divided into two groups: epirubicin at final concentrations of 4.0, 2.0, 1.0, 0.5 and 0.25 mg / L, respectively, Physiological saline. The expression of c-FLIP in the two cell lines was detected by RT-PCR and the percentage of apoptotic cells was detected by flow cytometry after 24, 48 and 72 hours. Results With the increase of epirubicin concentration and the prolongation of action time, the expression of c-FLIP decreased as a whole. With the increase of epirubicin concentration and the prolongation of action time, the apoptosis rate of breast cancer cells gradually increased , In a dose and time dependent manner. Among them, the apoptosis rate was the highest at 72 h at the concentration of 2 mg / L. Conclusion Epirubicin-induced apoptosis of breast cancer cells may be through inhibition of c-FLIP expression, and the effect of the concentration of 2 mg / L for 72 h was the best.
