目的 :探讨郑州地区汉族人群对氧磷酯酶 芳香酯酶 (Paraoxonase1 Arylesterase ,PON1 ArE) 192位基因多态性与 2型糖尿病 (DM)并发冠心病 (DM CHD)的关系。方法 :观察对象均为郑州地区汉族居民 ,其中DM组12 1例、DM CHD组 12 5例、非糖尿病急性心肌梗死 (AMI)组 5 0例、对照组 12 7例 ,用聚合酶链反应技术检测其PON1 ArE基因 192位多态性 ,终点法测定血清PON1 ArE活性 ,酶法与酶联免疫法测定胆固醇 (TC)、三酰甘油(TG)、高密度脂蛋白胆固醇 (HDL C)、低密度脂蛋白胆固醇 (LDL C)、氧修饰低密度脂蛋白 (ox LDL)等指标后 ,分析研究。结果 :①郑州地区汉族人群PON1 ArE192位Q、R基因频率 ,DM组为 0 .5 2、0 .4 8,DM CHD组为 0 .37、0 .6 3,AMI组 0 .5 3、0 .4 7,对照组分别为 0 .5 4、0 .4 6。DM与AMI组Q、R基因频率和对照组差异无显著性意义 ,DM CHD组R基因频率大于这 3组。②DM和DM CHD组PON1 ArE活性降低 ,降低幅度DM CHD组接近于AMI组 ,大于DM组。DM与DM CHD组内各基因型组中RR基因型患者ox LDL、TC、TG、载脂蛋白B1 0 0 等水平均高于QQ型 (P <0 .0 1) ,HDL C低于QQ型患者 ,DM CHD组中的RR基因型的异常改变最为明显。AMI组内各基因型的ox LDL无明显差异。结论 :郑州地区汉族人群存在PON1 ArE192位基因
Objective: To investigate the association between paraoxonase1 Arylesterase (PON1 ArE) 192 gene polymorphism and type 2 diabetes mellitus (DM) with coronary heart disease (CHD) in Han nationality in Zhengzhou area. Methods: The subjects were all Han nationality inhabitants in Zhengzhou, including 121 cases in DM group, 125 cases in DM CHD group, 50 cases in non-diabetic AMI group and 127 cases in control group. Polymerase chain reaction The polymorphism of PON1 ArE gene at position 192 was detected by enzyme linked immunosorbent assay (ELISA). Serum PON1 ArE activity was measured by end point method. The levels of cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL C) Density lipoprotein cholesterol (LDL C), oxygen-modified low density lipoprotein (ox LDL) and other indicators, the analysis and research. Results: ①The Q and R gene frequencies of PON1 ArE192 in Han nationality in Zhengzhou area were 0.55 and 0.848 in DM group, 0.37 and 6.36 in DM CHD group, .4 7, the control group were 0.54,0.46 respectively. The frequencies of Q and R genes in DM and AMI group were not significantly different from those in control group. The frequency of R gene in DM CHD group was higher than those in DM group. ② PON1 ArE activity was decreased and decreased in DM and DM CHD groups. Compared with DM group, DM CHD group was close to AMI group. The levels of ox LDL, TC, TG, apolipoprotein B1 0 0 in RR genotypes among DM genotype group and DM genotype group were higher than that of QQ genotype (P <0.01), HDL C was lower than that of QQ genotype Patients, DM CHD group RR genotype abnormal changes most obvious. There was no significant difference in the ox LDL between genotypes in AMI group. Conclusion: PON1 ArE192 gene exists in Han population in Zhengzhou area
