目的:探讨六味地黄丸对2型糖尿病(T2DM)代谢的影响及其作用机制。方法:取60只大鼠随机分为正常对照组、高脂(模型)组、模型+盐酸吡格列酮(降糖)治疗组、模型+六味地黄丸(试药)治疗35g/kg、模型+六味地黄丸(试药)治疗25g/kg、模型+六味地黄丸(试药)治疗15g/kg、高脂饲料伴链佐霉素制备T2DM模型。采用全自动生化分析仪测定血糖含量;酶联免疫吸附分析(ELISA)法测定血清脂联素、胰岛素水平;计算胰岛素敏感指数(ISI)及胰岛素抵抗(HOMA-IR)指数。结果:与模型组比较,六味地黄丸35g/kg、25g/kg、15g/kg剂量组均具有降低T2DM模型大鼠空腹血糖作用,使血清脂联素含量升高,调节胰岛素水平,ISI、HOMA-IR指数均具有非常显著的差异。结论:六味地黄丸具有调节病理状态下血糖、脂联素、胰岛素异常的作用,使机体对胰岛素敏感性提高,降低HOMA-IR,提示六味地黄丸有一定治疗T2DM。
Objective: To investigate the effect of Liuweidihuangwan on the metabolism of type 2 diabetes mellitus (T2DM) and its mechanism. Methods: 60 rats were randomly divided into normal control group, high fat (model) group, model + pioglitazone hydrochloride hypoglycemic group and model + Liuweidihuangwan (test drug) for 35g / kg, Pills (test) treatment 25g / kg, model + Liu Wei Di Huang Wan (test) treatment 15g / kg, high fat diet with streptozotocin preparation T2DM model. Serum adiponectin and insulin levels were measured by enzyme-linked immunosorbent assay (ELISA). Index of insulin sensitivity index (ISI) and insulin resistance (HOMA-IR) were calculated by automatic biochemistry analyzer. Results: Compared with the model group, the 35g / kg, 25g / kg and 15g / kg dose groups of Liuweidihuang Pill all reduced the fasting blood glucose in T2DM rats, increased the content of serum adiponectin, regulated the level of insulin, ISI, HOMA -IR index has a very significant difference. Conclusion: Liuweidihuangwan can regulate the blood glucose, adiponectin and insulin in pathological condition, improve the body’s insulin sensitivity and reduce HOMA-IR, suggesting that Liuweidihuangwan can treat T2DM.
