目的:观察孤儿核受体神经生长诱导基因B-β(nerve growth factor-induced gene B-β,NGFIB-β)在大鼠脊髓发育过程中蛋白表达的动态变化及与增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)的相关关系,探讨NGFIB-β在神经细胞迁移分化中的表达意义。方法:采用石蜡包埋组织切片,免疫组织化学染色及免疫组织化学双重染色。结果:NGFIB-β阳性表达在脊髓的发育过程中呈现出显著的动态变化,NGFIB-β阳性细胞显示已经分化的形态。NGFIB-β首先在胚胎13天(E13)脊髓组织的背侧出现阳性细胞,E15可清楚观察到阳性细胞呈“扶梯”样迁移方式从背侧向腹侧迁移,随着发育和细胞的成熟,阳性表达明显减少,成熟脊髓组织中未见阳性表达,与PCNA的对比观察表明,两种蛋白分别表达在不同的细胞中,NGFIB-β表达在迁移分化的幼稚神经细胞中,而在有增殖能力的细胞中不表达。结论:NGFIB-β可能在大鼠脊髓神经细胞从幼稚到成熟的迁移分化过程中具有调控作用。
OBJECTIVE: To observe the dynamic changes of protein expression of nerve growth factor-induced gene B-β (NGFIB-β) in orbital spinal cord of orphans and its relationship with proliferating cell nuclear antigen, PCNA) in order to explore the significance of NGFIB-β expression in the migration and differentiation of neural cells. Methods: Paraffin embedded sections, immunohistochemical staining and immunohistochemistry double staining. Results: The positive expression of NGFIB-β showed a significant dynamic change during the development of the spinal cord, and the NGFIB-β positive cells showed the differentiated morphology. NGFIB-β first showed positive cells on the dorsal side of spinal cord tissue on embryonic day 13 (E13). Positive cells were observed as “escalator” -like migrating pattern from the dorsal side to the ventral side on E15, Mature, positive expression was significantly reduced, no positive expression in the mature spinal cord tissue, and PCNA comparative observation showed that the two proteins were expressed in different cells, NGFIB-β expression in the migration of differentiated naive nerve cells, and in the presence of Proliferating cells were not expressed. Conclusion: NGFIB-β may play a regulatory role in the migration and differentiation of rat spinal cord neurons from naive to mature.
