目的:探讨黄荆子木脂素VBE-6联合顺铂对体外培养人卵巢癌CoC1细胞增殖的抑制作用。方法:将CoC1细胞培养并分别单独加入不同浓度VBE-6(0.1、1.0、5.0、10.0、20.0μmol/l)、顺铂(10.0μmol/l)单独及联合作用后,用MTT法检测卵巢癌CoC1细胞增殖情况,流式细胞术检测细胞凋亡率和细胞周期分布。结果:MTT法检测结果显示,不同浓度VBE-6组随着作用时间延长及药物浓度增加,CoC1细胞的增殖抑制率呈明显上升趋势,各组间比较,差异有统计学意义(P<0.05);联合用药组明显高于单一用药组,差异有统计学意义(P<0.05)。流式细胞术结果显示,VBE-6能阻滞卵巢癌CoC1细胞于G1期,同时可诱导细胞凋亡发生;随着VBE-6用药浓度增加,细胞凋亡率增高。结论:VBE-6对卵巢癌CoC1细胞具有增殖抑制和诱导凋亡作用,在一定浓度范围内,这种作用呈剂量-时间依赖性;VBE-6与顺铂联合应用具有协同作用。
OBJECTIVE: To investigate the inhibitory effect of Vitexin VBE-6 combined with cisplatin on the proliferation of human ovarian cancer CoC1 cells in vitro. Methods: CoC1 cells were cultured and treated with different concentrations of VBE-6 (0.1,1.0,5.0,10.0,20.0μmol / l) and cisplatin (10.0μmol / l) alone and in combination. MTT assay was used to detect ovarian cancer CoC1 cell proliferation, flow cytometry to detect apoptosis rate and cell cycle distribution. Results: The results of MTT assay showed that the proliferation inhibition rate of CoC1 cells in VBE-6 group increased significantly with the prolongation of action time and drug concentration, and the difference was statistically significant (P <0.05) ; Combination group was significantly higher than the single drug group, the difference was statistically significant (P <0.05). Flow cytometry results showed that VBE-6 could block the G1 phase of ovarian cancer CoC1 cells and induce apoptosis at the same time. As the concentration of VBE-6 increased, the apoptosis rate of cells increased. CONCLUSION: VBE-6 can inhibit proliferation and induce apoptosis of CoC1 cells in a dose-dependent manner. VBE-6 has a synergistic effect with cisplatin.
