目的:探讨ATR基因的表达对HeLa细胞对烷化剂敏感性的影响。方法:采用RNA干扰技术抑制HeLa细胞ATR基因的表达,观察ATR被阻断后HeLa细胞对烷化剂敏感性的变化,从而确定ATR基因的作用。结果:筛选到表达针对ATR基因的短发夹RNA(shRNA)阳性克隆,Western印迹结果显示ATR基因表达受到明显抑制;HeLa细胞对烷化剂的敏感性实验提示,ATRshRNA+HeLa细胞对烷化剂的敏感性明显增强。结论:抑制ATR基因可明显提高HeLa细胞对烷化剂的敏感性。
Objective: To investigate the effect of ATR gene expression on alkylating agent sensitivity in HeLa cells. METHODS: RNA interference was used to inhibit the expression of ATR gene in HeLa cells. The sensitivity of HeLa cells to alkylating agents after ATR block was observed to determine the role of ATR gene. Results: The shRNA-positive clones expressing ATR gene were screened by Western blot. The results of Western blotting showed that ATR gene expression was significantly inhibited. The sensitivity of HeLa cells to alkylating agents indicated that ATRshRNA + HeLa cells responded to alkylating agents The sensitivity is significantly enhanced. Conclusion: Inhibition of ATR gene can significantly increase the sensitivity of HeLa cells to alkylating agents.
