目的在现有方法的基础上进一步优化各种条件,建立简便、快速、准确的能同时测定麻黄中盐酸麻黄碱及盐酸伪麻黄碱含量的高效液相色谱法。方法色谱柱:Diamonsil-C18(250mm×4.6mm,5μm);流动相:乙腈-0.2%磷酸-三乙胺(3.8:96:0.2);流速:1ml.min-1;柱温:30℃;检测波长:207nm;进样量:20μl。结果盐酸麻黄碱、盐酸伪麻黄碱质量浓度线性范围分别为104.3μgml-1~521.5μg·ml-1(n=12,r=1),101.1μg·ml-1~505.5μg·ml-1(n=12,r=1),平均加样回收率分别为103.14%(n=6,RSD=0.76%和102.98%)(n=6,RSD=0.98%)。结论本法能同时测定麻黄中麻黄碱和伪麻黄碱两种成分的含量,快速准确,重现性好,优于药典法,可用于麻黄药材的质量控制。
OBJECTIVE To further optimize various conditions on the basis of the existing methods and to establish a simple, rapid and accurate HPLC method for simultaneous determination of ephedrine hydrochloride and pseudoephedrine hydrochloride in Ephedra. Method Column: Diamonsil-C18 (250mm × 4.6mm, 5μm); mobile phase: acetonitrile-0.2% phosphoric acid- triethylamine (3.8: 96: 0.2); flow rate: 1ml.min-1; Detection wavelength: 207nm; injection volume: 20μl. Results The linear ranges of ephedrine hydrochloride and pseudoephedrine hydrochloride were 104.3μgml -1 to 521.5μg · ml -1 (n = 12, r = 1) and 101.1μg · ml -1 to 505.5μg · ml -1 (n = 12, r = 1). The average recoveries were 103.14% (n = 6, RSD = 0.76% and 102.98%, respectively) with n = 6 and RSD = 0.98%. Conclusion The method can simultaneously determine the content of ephedrine and pseudoephedrine in ephedra, which is rapid, accurate and reproducible. It is superior to the pharmacopoeia and can be used for the quality control of ephedra.
