目的:对辣椒碱立方液晶凝胶进行表征,建立辣椒碱高效液相含量测定方法。方法:以甘油单油酸酯为液晶材料、辣椒碱为主药,采用自乳化方法制备了辣椒碱立方液晶凝胶;用偏光显微镜及小角衍射对立方液晶凝胶进行了表征;采用高效液相测定了立方液晶凝胶中辣椒碱的含量。结果:立方液晶凝胶在偏光显微镜下视野黑暗;小角衍射显示在0.109 6,0.133 4,0.155 7,0.188 3-1处出现信号峰,峰位置符合q1∶q2∶q3∶q4=2∶3∶4∶6,特征峰显示其内部结构为Pn3m(Q224)。高效液相色谱法测定立方液晶凝胶中的辣椒碱线性范围为3.25×10-4~2.08×10-2g.L-1(R2=1),平均回收率97.53%,RSD 2.9%(n=9)。结论:偏光显微镜及小角衍射适宜于立方液晶凝胶的表征,高效液相测定方法简便、可行。
OBJECTIVE: To characterize capsaicin cubic liquid crystal gel and establish a HPLC method for the determination of capsaicin high-performance liquid phase. Methods: Capsaicin cubic liquid crystal gel was prepared by self-emulsifying method using glycerol monooleate as liquid crystal material and capsaicin as main drug. The cubic liquid crystal gel was characterized by polarized light microscopy and small angle diffraction. The content of capsaicin in the cubic liquid crystal gel was measured. Results: The cubic liquid crystal gel showed a dark field of vision under a polarizing microscope. Small angle diffraction showed a signal peak at 0.109 6, 0.133 4, 0.1555 7 and 0.188 3-1. The peak position was in accordance with q1: q2: q3: q4 = 4: 6, the characteristic peak shows that its internal structure is Pn3m (Q224). The linear range was 3.25 × 10-4 ~ 2.08 × 10-2g.L-1 (R2 = 1). The average recoveries were 97.53% and RSD 2.9% (n = 9). Conclusion: Polarization microscopy and small angle diffraction are suitable for the characterization of cubic liquid crystal gel. The method of high performance liquid chromatography is simple and feasible.
