目的通过检测FHIT蛋白和WWOX蛋白在胃腺癌组织中表达情况和FHITmRNA和WWOXmRNA在胃腺癌和癌旁正常组织的表达情况,分析两者与临床病理特征之间的关系及两者之间是否具有相关性。方法随机抽取佳木斯大学附属第一医院普外科2000年到2011年期间的70例胃腺癌切除组织标本,采用免疫组化和RT-PCR方法检测胃腺癌组织中临床病理关系及胃腺癌与癌旁正常组织(距肿瘤边缘>5CM)的关系。结果 (1)FHIT和WWOX均阳性表达28例。FHIT和WWOX均阴性表达24例。FHIT阳性而WWOX阴性8例。FHIT阴性而WWOX阳性10例。FHIT和WWOX在胃腺癌中的表达成正相关性。(2)FHITmRNA与WWOXmRNA在70例胃腺癌组织中表达较癌旁组织低者分别有22例和28例;较癌旁胃正常组织高者均有6例。FHITmRNA和WWOXmRNA在胃腺癌的表达低于癌旁胃组织。结论 (1)FHIT和WWOX在胃腺癌组织中阳性表达率较低,在组织学分级,病理分期中呈正相关,而与年龄和病理分型无相关性。(2)FHIT和WWOX的在胃腺癌组织中与癌旁胃正常组织中差异有显著性。(3)FHITmRNA和WWOXmRNA在胃腺癌组织中的低表达呈低度正相关性。(4)FHIT和WWOX是重要的抑癌基因。
OBJECTIVE: To detect the expression of FHIT protein and WWOX protein in gastric adenocarcinoma and the expression of FHIT mRNA and WWOX mRNA in gastric adenocarcinoma and adjacent normal tissues, and to analyze the relationship between them and clinicopathological features and whether they are related Sex. Methods Seventy patients with gastric adenocarcinoma resected from 2000 to 2011 in the Department of General Surgery, the First Affiliated Hospital of Jiamusi University were enrolled in this study. The clinicopathological features, gastric adenocarcinoma and adjacent normal tissues were detected by immunohistochemistry and RT-PCR Tissue (away from tumor margin> 5 cm). Results (1) FHIT and WWOX were positive in 28 cases. FHIT and WWOX were negative in 24 cases. FHIT positive and WWOX negative in 8 cases. FHIT negative and WWOX positive in 10 cases. FHIT and WWOX expression in gastric adenocarcinoma has a positive correlation. (2) The expression of FHIT mRNA and WWOX mRNA in 70 cases of gastric adenocarcinoma tissues were lower than those in the paracancerous tissues, 22 cases and 28 cases, respectively; 6 cases were higher than normal gastric tissue. The expression of FHIT mRNA and WWOX mRNA in gastric adenocarcinoma was lower than that in adjacent gastric tissues. Conclusions (1) The positive rates of FHIT and WWOX are lower in gastric adenocarcinoma tissues, positive in histological grade and pathological stage, but not with age and pathological type. (2) There was significant difference between FHIT and WWOX in gastric adenocarcinoma tissues and adjacent normal gastric tissues. (3) Low expression of FHIT mRNA and WWOX mRNA in gastric adenocarcinoma showed a low positive correlation. (4) FHIT and WWOX are important tumor suppressor genes.
