探讨人参皂甙(GS)对人骨髓CD34+造血干细胞的增殖作用。方法:采给Dynal M450 CD34+免疫磁珠分离法阳性选择获取高纯度的人骨髓CS34+造血干细胞,应用多向祖细胞(CFU-Mix)体外培养技术,观察CD34+造血干细胞对GS的敏感性。结果:1.经Dynal M-450 CD34+免疫磁珠分离获得骨3髓 CD34+细胞占骨髓单个核细胞( MNC)的 0. 52~1.30%;CD34+细胞的存活率达94.0~98.2%。 2最佳浓度的GS使CD34+细胞的CFU-Mix集落形成率提高54.4%(P<0.01),而高浓度时表现为抑制。结论:GS不但作用于人骨髓造血祖细胞,而已对CD34+造血干细胞也具有明显的刺激增殖作用。
To investigate the effect of ginsenosides (GS) on the proliferation of human bone marrow CD34 + hematopoietic stem cells. METHODS: Human bone marrow CS34 + hematopoietic stem cells with high purity were positively selected by Dynal M450 CD34+ immunomagnetic bead separation method. The sensitivity of CD34 + hematopoietic stem cells to GS was observed by in vitro culture of multi-progenitor cells (CFU-Mix). Results: 1. Bone marrow 3 marrow CD34+ cells were isolated from bone marrow mononuclear cells (MNC) by Dynal M-450 CD34+ immunomagnetic beads. 52 to 1.30%; the survival rate of CD34+ cells reached 94.0 to 98.2%. 2The optimal concentration of GS increased the CFU-Mix colony formation rate of CD34+ cells by 54.4% (P<0.01), but showed inhibition at high concentrations. CONCLUSION: GS not only acts on human bone marrow hematopoietic progenitor cells, but also has significant stimulation on CD34+ hematopoietic stem cells.
