目的了解江西省流行性腮腺炎病毒(MuV)的基本特征。方法采集腮腺炎患者咽拭子标本分离病毒,对分离到的MuV小疏水蛋白(small hydrophobic protein,SH)基因316个核苷酸片段进行逆转录PCR(RT-PCR)扩增,对扩增产物进行序列测定及基因分型。结果江西省27株Mu V分离株属于F基因型,核苷酸及氨基酸同源性分别为94.6%~100%和89.5%~100%。与F基因型参考株序列相比,与兰州株的氨基酸同源性为91.2%~94.7%,与上海株的氨基酸同源性为84.2%~87.7%。Mu V的F基因型毒株呈现特异性突变(C~(Nt65)、C~(Nt105)、G~(Nt137)、C~(Nt192)、C~(Nt239));在与基因分型有关的氨基酸三联体上,有1株为VML,1株为ILL,其余均为IML。结论江西省目前腮腺炎病毒流行株优势基因型为F基因型。现有的F基因型Mu V流行株与A基因型疫苗株S79差异较大。氨基酸三联体已出现变异,因此需继续加强本省腮腺炎病毒监测的基因型监测。
Objective To understand the basic characteristics of mumps virus (MuV) in Jiangxi Province. Methods The virus was isolated from the pharyngeal swab specimens of mumps and the 316 bp fragment of Mu hydrophobic small hydrophobic protein (SH) gene was amplified by reverse transcription polymerase chain reaction (RT-PCR). The amplification products Sequencing and genotyping were performed. Results The 27 strains of Mu V isolated from Jiangxi Province were F genotypes with nucleotide and amino acid identities of 94.6% -100% and 89.5% -100%, respectively. The amino acid homology with Lanzhou strain was 91.2% ~ 94.7% compared with the F genotype reference strain, and the amino acid homology with Shanghai strain was 84.2% ~ 87.7%. The F genotypes of Mu V showed specific mutations (C ~ (Nt65), C ~ (Nt105), G ~ (Nt137), C ~ (Nt192), C ~ (Nt239) Of the amino acid triplet, a strain of VML, a strain of ILL, the rest are IML. Conclusion The predominant genotype of mumps virus in Jiangxi Province is F genotype. The existing F genotype Mu V epidemic strain and A genotype vaccine strain S79 differences. Amino acid triplets have mutated, so the need to continue to strengthen the mumps virus monitoring of genotype monitoring.
