目的:研究桂苓神术汤的利尿作用,筛选利尿作用的有效部位并探讨其作用机制。方法:SD大鼠,随机分为18组:正常组、模型组,呋噻米组,桂苓神术汤及石油醚部位、乙酸乙酯部位、正丁醇部位、剩余部位分别设10.0,5.0,2.5 g.kg-1 3个剂量组。每组大鼠均按40 mL.kg-1生理盐水ig,形成盐水负荷模型。45 min后,ig给予不同剂量的桂苓神术汤及不同极性部位,用大鼠代谢笼法观察大鼠6 h的尿量,并测定尿液pH及Na+,K+,Cl-的浓度。结果:桂苓神术汤组(5.0,2.5 g.kg-1),乙酸乙酯部位组(5.0 g.kg-1),剩余部位组(5.0 g.kg-1)均能显著增加盐水负荷大鼠6 h的尿量;尿中K+排出量较模型组均显著升高,分别为(850.3±78.8),(816.6±91.1),(826.3±84.9),(776.7±69.1)mmol.L-1,Na+,Cl-排出量较模型组无显著差异。结论:桂苓神术汤及有效部位具有缓和持久的利尿作用,其利尿作用机制可能为增加大鼠尿液中K+的排泄。
Objective: To study the diuretic effect of Guiling Decoction, to screen the effective site of diuretic effect and to explore its mechanism. Methods: SD rats were randomly divided into 18 groups: normal group, model group, furosemide group, Guiling Shenzhu decoction and petroleum ether, ethyl acetate and n-butanol, and the remaining sites were set at 10.0, 5.0 , 2.5 g.kg-1 3 dose groups. Each group of rats according to 40 mL.kg-1 saline ig, the formation of saline load model. After 45 min, different doses of Guiling Decoction and different polarity sites were given to ig. Urinary output of 6 h after rats was observed by metabolic cages in rats. Urine pH, Na +, K + and Cl- concentrations were also measured. Results: Guilin Ling Shu Decoction group (5.0,2.5 g.kg-1), ethyl acetate group (5.0 g.kg-1), the rest of the group (5.0 g.kg-1) can significantly increase the saline load The urinary excretion of K + in rats was significantly higher than that of the model group (850.3 ± 78.8), (816.6 ± 91.1), (826.3 ± 84.9) and (776.7 ± 69.1) mmol.L- 1, Na +, Cl-discharge than the model group no significant difference. Conclusion: Guiling Decoction and its effective site have a moderate and long-lasting diuretic effect, and its diuretic mechanism may be to increase the excretion of K + in the urine of rats.
