目的观察针箭颗粒对自发性高血压(SHR)大鼠左室肥厚(LVH)的治疗作用并探讨其可能机理。方法取13周龄SHR大鼠40只随机分为针箭颗粒高、中、低剂量治疗组与卡托普利组及模型对照组五组,另取WKY大鼠6只为正常对照组;各组实验8周后进行血压、左室重与体重比(LVM/BW)及循环和局部血管紧张素II(AngII)浓度(放免法测定)的组间比较。结果21周龄时SHR大鼠LVM/BW显著高于WKY大鼠,治疗后各治疗组血压均显著下降,针箭颗粒高、中剂量组及卡托普利组LVM/BW、心肌局部AngII浓度均明显降低;LVM/BW与血压及循环AngII无明显相关,而与心肌局部AngII明显正相关。结论针箭颗粒降低SHR大鼠血压的同时显著逆转其LVH,其作用机理可能主要与降低心肌局部肾素血管紧张素系统(RAS)活性有关。
Objective To observe the therapeutic effect of Zhujian Granules on left ventricular hypertrophy (LVH) in spontaneously hypertensive rats and explore its possible mechanism. Methods Thirty-four SHR rats aged 13 weeks were randomly divided into high-, middle-, and low-dose treatment group of Qijian granules, five groups of captopril group and model control group, and another 6 WKY rats were taken as normal control group. Eight weeks after the group experiment, blood pressure, left ventricular mass to body weight ratio (LVM/BW) and circulating and local angiotensin II (Ang II) concentrations (radioimmunoassay) were compared between groups. Results The LVM/BW of SHR rats was significantly higher than that of WKY rats at 21 weeks of age. The blood pressure of each treatment group was significantly decreased after treatment, and the concentration of LVM/BW and myocardial Ang II in the high- and middle-dose group of the Zhujian granules and captopril group. All significantly decreased; LVM/BW was not significantly associated with blood pressure and circulating AngII, but was positively correlated with local AngII in the myocardium. Conclusion Acupuncture granules can significantly reduce blood pressure in SHR rats while reversing LVH. The mechanism may be related to the reduction of local renin angiotensin system (RAS) activity.
