目的探讨血管内皮生长因子C及其受体VEGF-R3(fms-liketyrosinekinase-4,Flt-4)在人直肠腺癌淋巴管生成中的作用。方法用免疫组化SABC染色法观察32例人直肠腺癌组织中VEGF-C及其受体VEGF-R3(fms-liketyrosinekinase-4,Flt-4)和Podoplanin表达的情况。结果直肠腺癌组织周边部淋巴管密度比内部明显增高(P<0.01);直肠腺癌组织周边部VEGF-C及其受体VEGF-R3阳性细胞数比内部明显增多(P<0.01)。VEGF-C及其受体VEGF-R3表达阳性细胞密度和淋巴管密度呈正相关;直肠腺癌组织中VEGF-C及其受体VEGF-R3异位表达在淋巴管内皮细胞上,而在血管中未见表达,差异有显著性(P<0.01)。结论VEGF-C及其受体VEGF-R3促进直肠腺癌淋巴管的病理生成,淋巴管生成提示在为肿瘤细胞提供养分的同时,也为肿瘤转移创造了条件。
Objective To investigate the role of vascular endothelial growth factor C and its receptor VEGF-R3 (fms-liketyrosinekinase-4, Flt-4) in lymphangiogenesis of human rectal adenocarcinoma. Methods Immunohistochemical SABC staining was used to observe the expression of VEGF-C, VEGF-R3 (fms-liketyrosinekinase-4, Flt-4) and Podoplanin in 32 human rectal adenocarcinoma. Results The density of lymphatic vessels around the rectal adenocarcinoma was significantly higher than that inside (P <0.01). The number of positive cells of VEGF-C and its receptor VEGF-R3 in the rectal adenocarcinoma tissues was significantly increased (P <0.01). VEGF-C and its receptor VEGF-R3 expression positive cell density and lymphatic vessel density was positively correlated; rectal adenocarcinoma VEGF-C and its receptor VEGF-R3 ectopic expression in lymphatic endothelial cells, and in the blood vessels No expression, the difference was significant (P <0.01). Conclusion VEGF-C and its receptor VEGF-R3 promote lymphangiogenesis in rectal adenocarcinoma. Lymphangiogenesis suggests that while providing nutrients for tumor cells, it also creates a condition for tumor metastasis.
