目的探讨去势法联合皮质激素快速诱导骨质疏松(OP)模型兔的可行性。材料与方法 40只8月龄雌性大白兔随机分为假手术组(A组,n=20)及去势+肌注甲强龙组(B组,n=20)。术后第2、4、8、10周行血清骨代谢标志物检测及离体L5椎体显微CT(Micro-CT)扫描。结果两组骨密度(BMD)在OVX术后第8周开始差异有统计学意义,而骨体积分数(BV/TV)、骨小梁连接密度(Conn.D)、骨小梁数目(Tb.N)、结构模型指数(SMI)、骨小梁间隙(Tb.Sp)及骨小梁厚度(Tb.Th)分别在术后4、8周开始差异有统计学意义。结论去势联合肌注甲强龙可在8周内快速建立OP兔模型,骨微结构参数变化早于BMD降低。
Objective To investigate the feasibility of using castration combined with corticosteroids to induce osteoporosis (OP) model rabbits rapidly. MATERIALS AND METHODS Forty eight-month-old female white rabbits were randomly divided into sham operation group (n = 20) and castration + intramuscular injection of methylprednisolone group (n = 20). At 2, 4, 8, and 10 weeks after operation, serum markers of bone metabolism and Micro-CT scanning of L5 vertebral body were performed. Results The bone mineral density (BMD) of the two groups had statistical significance at the beginning of the 8th week after OVX, while the ratio of BV / TV, Conn.D and Tb. N), SMI, Tb.Sp, and Tb.Th were significantly different at 4 and 8 weeks after operation. Conclusion Ovariectomy combined with intramuscular injection of methylprednisolone can rapidly establish OP rabbit model within 8 weeks, and the change of microarchitectural parameters is earlier than that of BMD.
