目的 研究延迟整流性 K+ 通道在肿瘤坏死因子α( T N Fα) 抑制人肝癌细胞增殖中的作用。方法 采用膜片钳技术的全细胞记录方式记录人肝癌细胞的跨膜电流,氚标记的胸腺嘧啶核苷掺入技术测定人肝癌细胞的 D N A 合成能力。结果 T N Fα抑制肝癌细胞增殖的同时,未能使肝癌细胞膜离子通道的跨膜 K+ 电流和细胞膜电导降低。 K+ 通道阻断剂四乙胺不能进一步增加 T N F 抑制 D N A 合成的作用,且无明显量效关系和时间依赖性。结论 T N Fα抑制人肝癌细胞增殖不是通过对细胞膜 K+通道的干扰来实现。
Objective To study the role of delayed rectifier K+ channels in inhibiting the proliferation of human hepatoma cells by tumor necrosis factor alpha (T N Fα). Methods Transmembrane currents of human hepatoma cells were recorded by whole-cell recording using patch-clamp technique, and D N A synthesis capacity of human hepatoma cells was measured by tritiated thymidine incorporation technique. Results T N F α inhibited the proliferation of hepatocellular carcinoma cells and failed to decrease the transmembrane K + current and membrane conductance of the membrane channel of hepatoma cells. K+ channel blocker tetraethylamine could not further increase the effect of T N F on the inhibition of D N A synthesis, and there was no obvious dose-effect relationship and time dependence. Conclusion The inhibition of proliferation of human hepatocellular carcinoma cells by T N F α is not achieved by interference with the cell membrane K + channels.
