根据实验室测序的大豆线粒体基因组片段设计线粒体细胞色素b(Cytochrome,cob)基因特异引物,PCR扩增获得1 173 bp大豆(JLGM-1B)线粒体cob基因保守区序列。经序列分析,该基因编码391个氨基酸,A+T含量58.1%,G+C含量41.9%。Southern杂交表明cob基因在栽培大豆线粒体基因组中至少有2个拷贝。氨基酸序列及系统进化树分析表明,大豆与其它植物的cob基因具有较高的同源性,与双子叶作物同源性高达98%,其分类地位与传统系统分类地位相吻合,说明cob基因具有较高的保守性,可以作为分子系统分类学潜在的遗传分化标记。
The conserved region of mitochondrial cob gene of 1 173 bp soybeans (JLGM-1B) was obtained by PCR amplification of mitochondrial cytochrome b (bbc) specific primers based on the laboratory sequencing of soybean mitochondrial genome. After sequence analysis, the gene encoding 391 amino acids, A + T content of 58.1%, G + C content of 41.9%. Southern blotting showed that the cob gene has at least two copies in the genome of cultivated soybean mitochondria. Amino acid sequence analysis and phylogenetic tree analysis showed that the cob gene of soybean and other plants had high homology with 98% homology with dicotyledonous crops. The taxonomic status of cob gene was in accordance with the traditional taxonomic status, indicating that cob gene has Higher conservatism can be used as a molecular taxonomy for potential genetic markers of differentiation.
