目的:探讨左旋棉酚对人纤维肉瘤细胞HT1080和人皮肤成纤维细胞HSF的抑制增殖作用。方法:采用MTT法检测药物对细胞增殖的抑制率,在倒置显微镜下观察细胞的形态学变化。结果:左旋棉酚对HT1080细胞和HSF细胞均有抑制增殖作用,呈浓度与时间依赖性。左旋棉酚对HT1080细胞作用24、48、72、96小时的IC_(50)分别为56.6、12.6、9.2、7.0μM,而对HSF细胞作用24、48、72、96小时的IC_(50)分别为192.3、53.6、42.5、19.4μM。随着药物浓度的增加,HT1080细胞数逐渐较少,脱落的细胞逐渐增多。而HSF细胞变化不明显。结论:左旋棉酚对人纤维肉瘤HT1080细胞有明显抑制增殖和选择性杀伤作用。
Objective: To investigate the inhibitory effect of L-Gossypol on the proliferation of human fibrosarcoma cell line HT1080 and human dermal fibroblast HSF. Methods: The inhibitory rate of drug on cell proliferation was detected by MTT assay. The morphological changes of cells were observed under inverted microscope. RESULTS: L-Gossypol could inhibit the proliferation of HT1080 cells and HSF cells both in concentration-and time-dependent manner. IC 50 of 24,48,72,96 hours after exposure to L-gossypol on HT1080 cells were 56.6, 12.6, 9.2 and 7.0 μM, respectively, whereas IC 50 of 24, 48, 72 and 96 hours 192.3, 53.6, 42.5, 19.4 μΜ. With the increase of drug concentration, the number of HT1080 cells gradually decreased, and the number of exfoliated cells gradually increased. HSF cells did not change significantly. Conclusion: L-gossypol can significantly inhibit the proliferation and selective killing of human fibrosarcoma HT1080 cells.
