为探讨实验性变态反应性神经炎(EAN) 中周围神经浸润CD4( + ) T细胞的凋亡,应用免疫组织化学和原位末端标记法动态检测不同发病阶段EAN 大鼠坐骨神经浸润CD4( + ) T 细胞的形态学改变及断裂的DNA 片段。结果发现具有凋亡形态学特征的浸润CD4( + ) T 细胞于免疫后第18 d 出现,在第22 d 达高峰后下降,第30 d 基本消失。原位末端标记法证实凋亡CD4( + ) T 细胞中存在断裂的DNA 片段。提示CD4( + ) T细胞凋亡是EAN 浸润CD4( + ) T细胞的重要清除机制
To investigate the apoptosis of peripheral blood infiltrating CD4 (+) T cells in experimental allergic neuritis (EAN), immunohistochemistry and in situ end-labeling were used to detect the invasiveness of sciatic nerve CD4 (+) in different stages of EAN. T cell morphological changes and broken DNA fragments. The results showed that infiltrating CD4 (+) T cells with morphological features of apoptosis appeared on the 18th day after immunization and reached the peak on the 22nd day, but disappeared on the 30th day. In situ terminal labeling confirmed the presence of fragmented DNA fragments in apoptotic CD4 (+) T cells. It is suggested that CD4 (+) T cell apoptosis is an important mechanism of clearance of EAN infiltrating CD4 (+) T cells
