本文建立了骆驼蓬碱的高效液相层析分析法,使用C_(18)柱反相层析,流动相为乙腈:水=70:30(V/V),加有枸橼酸0.05M,PH_4.0,流速0.8ml/min,以氨苯吖啶(mAMSA)为内标准,其保留时间为4.9~5.0min,骆驼蓬碱则为6.5min。 将上述方法应用于免血药浓度测定,数据经IBM—PC计算机处理,5兔中4兔药时曲线符合一室开放模型,仅一兔符合二室开放模型。各兔的药动学参数均经算出,其平均消除相半衰期为26.5±16.2min。
In this paper, a high performance liquid chromatography method was developed for the determination of harga in alkali. C 18 column reversed-phase chromatography was used. The mobile phase consisted of acetonitrile: water = 70:30 (V / V) PH_4.0, the flow rate of 0.8ml / min, with ammonia benzene acridine (mAMSA) as an internal standard, the retention time of 4.9 ~ 5.0min, compared to the harmalamin for 6.5min. The above method was applied to the determination of free blood drug concentration. The data were processed by IBM-PC computer. The curves of 4 rabbits in 5 rabbits accorded with the model of open chamber and only one rabbit accorded with 2-compartment open model. The pharmacokinetic parameters of each rabbit were calculated, the average elimination half-life was 26.5 ± 16.2min.
