用99mTc标记抗人活化血小板GMP-140MCAbSZ-51,对实验性狗股动脉和股静脉血栓模型进行了免疫显像研究.结果表明:血栓显示清晰,其最佳显像时间为注射99mTc-SZ-51后4~6小时;99mTc-SZ-51引血液半清除时间T1/2α=16.25±13.39分钟、T1/2β=7.28±1.15小时;注射99mTc-SZ-51后8小时处死狗,离体血栓/血液和血栓/周围肌肉单位质量放射性比值:动脉血栓为57.09±12.24和177.46±119.93,静脉血栓为6.43±4.70和94.46±57.59;99mTc标记非特异IgG,未见血栓显示.提示99mTc-SZ-51具有活体内导向定位血栓的能力,用于血栓性疾病的放免显像诊断有一定的可行性。
Immunohistochemical studies of experimental dog femoral and femoral vein thrombosis models were performed using 99mTc labeled anti-human activated platelet GMP-140MCAbSZ-51. The results showed that the thrombosis showed clear, the best imaging time was 4 ~ 6 hours after injection of 99mTc-SZ-51, the half-clearing time of 99mTc-SZ-51 blood was T1 / 2α = 16.25 ± 13.39 minutes, T1 /2β = 7.28 ± 1.15 hours; dogs were sacrificed 8 hours after injection of 99mTc-SZ-51, and the ratio of isolated thrombus / blood and thrombus / peripheral muscle mass radioactivity: arterial thrombus was 57.09 ± 12.24 and 177.46 ± 119.93, venous thrombosis was 6.43 ± 4.70 and 94.46 ± 57.59; 99mTc labeled non-specific IgG, no thrombus showed. It is suggested that 99mTc-SZ-51 has the ability of targeting thrombosis in vivo and it is feasible to diagnose thromboembolic disease by radioimmunoassay.
