多发性骨髓瘤(MM)是B淋巴细胞的恶性肿瘤。氧化砷(As_2O_3)已成功地用于急性早幼粒细胞白血病的治疗,其机制是诱导白血病细胞凋亡。我们初步研究了As_2O_3对MM细胞株(KM_3)的作用及其机制,旨在为MM的治疗提供借鉴。材料和方法1 试剂 As_2O_3(分析纯)溶于生理盐水,制成10~(-3)mol/L贮存液。工作浓度为0.5~6.0μmol/L。2 细胞培养 KM_3细胞由本室长期传代。以2×10~5/ml细胞浓度接种于含10%灭活FCS、100U/ml青霉素、100μg/ml链霉素的新鲜RPMI 1640培养液中培养(37℃,5%CO_2),每24小时用台盼蓝拒染法计数
Multiple myeloma (MM) is a malignant tumor of B lymphocytes. Arsenic oxide (As2O3) has been successfully used in the treatment of acute promyelocytic leukemia, and its mechanism is to induce apoptosis in leukemic cells. We preliminary studied the effect and mechanism of As_2O_3 on MM cell line (KM_3) in order to provide reference for the treatment of MM. Materials and Methods 1 Reagents As_2O_3 (analytically pure) was dissolved in physiological saline and made into a 10~(-3) mol/L stock solution. The working concentration is 0.5 to 6.0 μmol/L. 2 Cell culture KM_3 cells were passaged by the chamber for a long time. Inoculate 2×10 5 /ml cells in fresh RPMI 1640 culture medium containing 10% inactivated FCS, 100 U/ml penicillin, and 100 μg/ml streptomycin (37°C, 5% CO 2 ) every 24 hours.
