目的观察柴苓汤对慢性环孢素A肾病(chronic cyclosporine A nephropathy,CCN)大鼠一氧化氮(NO)、血管内皮素-1(ET-1)的影响。方法选取清洁级SD大鼠40只,随机分为对照组、模型组、缬沙坦组、柴苓汤组,每组10只。模型组、缬沙坦组及柴苓汤组采用经口灌服环孢素A[cyclosporine A,Cs A,30mg/(kg·d)]的方法制备慢性环孢素A肾病大鼠模型,对照组每日给予等容量橄榄油;造模的同时,缬沙坦组给予缬沙坦[10mg/(kg·d)],柴苓汤组给予柴苓汤[3g/(kg·d)],其余两组给予同剂量生理盐水灌胃,共28天,测定大鼠肾功能指标血清尿素氮(BUN)、血清肌酐(SCr)及肌酐清除率(CCr),检测血清中NO、ET-1的含量。结果与对照组比较,模型组大鼠肾功能明显下降(P<0.01),血清中NO含量显著降低(P<0.01),ET-1含量显著升高(P<0.01);经缬沙坦和柴苓汤干预后,肾功能显著提高(P<0.01),血清中NO含量显著上升(P<0.01),ET-1含量显著降低(P<0.01),且两组间比较无统计学意义。结论柴苓汤可改善CCN大鼠肾功能,调节NO、ET-1的表达,保护血管内皮功能,减轻肾损害。
Objective To observe the effect of Chailing Decoction on nitric oxide (NO) and endothelin-1 (ET-1) in rats with chronic cyclosporine A nephropathy (CCN). Methods Forty clean SD rats were randomly divided into control group, model group, valsartan group and Chailingtang group, with 10 rats in each group. The model group, Valsartan group and Chailingtang group were given chronic cyclosporin A nephropathy rat model by oral administration of cyclosporine A (CsA, 30mg / (kg · d)], control The rats in each group were treated with valsartan [10 mg / (kg · d)] at the same time as the model group and Chailing Decoction [3 g / (kg · d)], The remaining two groups were given the same dose of saline for 28 days. The serum levels of BUN, SCr and creatinine (CCr) were measured and the level of serum NO, ET-1 content. Results Compared with the control group, the renal function of the model group was significantly decreased (P <0.01), the content of NO in serum was significantly decreased (P <0.01) and the content of ET-1 was significantly increased (P <0.01) After the intervention of Chailing Decoction, the renal function was significantly increased (P <0.01), the content of NO in serum was significantly increased (P <0.01) and the content of ET-1 was significantly decreased (P <0.01), and there was no significant difference between the two groups. Conclusion Chailing Decoction can improve renal function, regulate the expression of NO and ET-1 in CCN rats, protect vascular endothelial function and reduce renal damage.
