目的:考察Ⅱ型ZTC1+1天然澄清剂在人参多糖提取中的纯化效果,并优化人参多糖的最佳纯化工艺。方法:采用星点设计-响应面法来安排实验,以多糖含量为指标,考察II型ZTC1+1天然澄清剂加入量、纯化时间、纯化温度对纯化效果的影响。结果:Ⅱ型ZTC1+1天然澄清剂纯化人参多糖的最佳工艺为:加入的澄清剂用量(B∶A=2∶1)为药液体积分数的2%,纯化温度为73℃,纯化时间135 min,此工艺条件下获得最大人参多糖保留率为67.9%。结论:II型ZTC1+1天然澄清剂能有效纯化人参多糖,且基于星点设计-响应面法优化的纯化工艺,方法简单可行,精密度高,预测性好。
OBJECTIVE: To investigate the purification effect of type Ⅱ ZTC1 + 1 natural clarifying agent on the extraction of polysaccharide from ginseng and optimize the purification process of polysaccharide from ginseng. Methods: The experimental design was carried out by the method of star point design - response surface methodology. Polysaccharide content was taken as an index to investigate the effect of ZTC1 + 1 type II natural clarifying agent, purification time and purification temperature on the purification effect. Results: The optimal process of purifying ginseng polysaccharides with type Ⅱ ZTC1 + 1 natural clarifier was as follows: the amount of clarifier added (B: A = 2:1) was 2% of the volume of the liquid, the purification temperature was 73 ℃, the purification time 135 min, the maximum ginseng polysaccharide retention rate of 67.9% under this process conditions. Conclusion: Type II ZTC1 + 1 natural clarifier can effectively purify ginseng polysaccharides. Based on the optimized design of star-point design-response surface method, the method is simple and feasible, high precision and good predictability.
