目的:探讨各种嗜酸细胞活化因子对支气管哮喘患者末梢血嗜酸细胞表面粘附分子表达的影响。方法:嗜酸细胞分离采用Percol不连续比重梯度离心法,粘附分子测定采用流式细胞仪FACScna。结果:血小板活化因子(PAF)、P物质(SP)、神经激肽A(NKA)、卟啉醇肉豆蔻酸乙酸酯(PMA)及粒—巨噬细胞集落刺激因子(GM—CSF)均具有增强嗜酸细胞表面粘附分子LFA1β及Macl表达的作用(P<0.01);而VLA4,LECAM—I于PAF、GM—CSF、PMA及fMLP刺激后明显降低(P<0.01),IL-4刺激后无变化。结论:PAF、SP、NKA、GM—CSF及PMA作为嗜酸细胞活化因子通过增强其表面粘附分子LFA1β及Macl的表达在支气管哮喘病理形成上发挥着重要作用
Objective: To investigate the effect of various eosinophil activation factors on the expression of adhesion molecules on the surface of eosinophils in patients with bronchial asthma. Methods: The eosinophils were separated by Percol discontinuous gravity gradient centrifugation and the adhesion molecules were determined by flow cytometry FACScna. Results: PAF, SP, NKA, PMA and GM-CSF (P <0.01). However, VLA4 and LECAM-I significantly decreased after stimulation with PAF, GM-CSF, PMA and fMLP (P <0.01), and increased the expression of LFA1β and Macl in eosinophils No change after IL-4 stimulation. CONCLUSION: PAF, SP, NKA, GM-CSF and PMA, as eosinophil activators, play an important role in the pathological development of bronchial asthma by enhancing the expression of their surface adhesion molecules LFA1β and Macl
