目的探讨降钙素基因相关肽((CGRP)及其拮抗剂(h-CGRP 8-37)与正常和应激情况下大鼠胃内胆汁返流的关系。方法 SD大鼠85只,实验分三部分:第一部分大鼠20只随机分为四组:对照组和CGRP低、中、高剂量组各5只,分别腹腔注射生理盐水(1 ml)和CGRP(10 μg/kg 1ml)、CGRP(30μg/kg 1 ml)和CGRP(1 ml),0.5 h后处死,取胃液测胆汁酸(TBA)浓度。第二部分采用冷束缚应激法,将35只大鼠分为两组,对照组5只,实验组30只,从浸入水中开始取2、4、5、6、8、10 h共6个时段,每个时段各5只,分别检测其胃内胆汁酸的浓度和胃黏膜的溃疡指数(用Guth评分),用放免试剂盒检测幽门区CGRP含量。第三部分大鼠30只随机分为两组:应激组和h-CGRP 8-37组各15只,从浸入水中开始取2、4、6 h共3个时段,每个时段各5只。取胃液测TBA浓度。结果正常大鼠在腹腔注射不同剂量CGRP 0.5 h后胃内胆汁酸浓度明显增高;应激性溃疡大鼠于应激结束后2h(即从应激开始计时6 h)胃内胆汁酸达到峰值,溃疡指数和幽门区CGRP含量分别于应激结束后4 h达到峰值和降到波谷;经幽门局部给予CGRP拮抗剂能显著降低应激性溃疡大鼠胃内胆汁返流程度。结论 CGRP能促进胃内肌汁酸反流,CGRP参与了幽门括约肌舒张功能的调控。

Gastric mucosa ulcer index (using Guth score), using radioimmunoassay kit detection of pyloric area CGRP content.Part three rats were randomly divided into two groups: the stress group and h-CGRP 8-37 group of 15, from The rats were immersed in water for 3, 4 and 6 h, respectively, with 5 rats in each time period.The TBA concentration in gastric juice was measured.Results The bile acid concentration in stomach was significantly increased after intraperitoneal injection of CGRP at different doses for 0.5 h Stress ulcer rats The bile acid peaked at 2 h after stress (ie, 6 h from the start of stress), and the ulcer index and the content of CGRP in pylorus peak reached the peak 4 h after the end of stress and dropped to the trough. The CGRP Antagonists can significantly reduce the degree of gastric bile reflux in stress ulcer rats.Conclusion CGRP can promote intramuscular acid reflux and CGRP is involved in the regulation of diastolic function in pyloric sphincter.
