目的:探索ras、myc基因蛋白在正常胎盘发育中的作用及ras、myc基因蛋白表达与葡萄胎临床预后关系。方法:用免疫组化方法,分析40例正常胎盘及50例葡萄胎组织H-ras和C-myc基因蛋白表达情况。结果:孕早期胎盘ras、myc基因蛋白的表达部位主要在细胞滋养细胞,表达率分别为70%和80%,孕中期表达部位主要在合体滋养细胞,表达率逐渐降低,足月时表达消失。葡萄胎ras、myc基因蛋白表达率分别为52%和60%,myc基因蛋白表达与滋养细胞的增生程度有关(P<001),ras、myc基因蛋白表达与葡萄胎恶变有关(分别P<001,P<005)。结论:ras、myc基因蛋白的过度表达与葡萄胎恶变有关。
Objective: To explore the role of ras and myc in the development of normal placenta and the relationship between ras and myc gene expression and the prognosis of hydatidiform mole. Methods: The expressions of H-ras and C-myc proteins in 40 normal placentae and 50 cases of hydatidiform mole were analyzed by immunohistochemistry. Results: The placental expression of ras and myc in the first trimester of pregnancy was mainly in the cytotrophoblast, the expression rates were 70% and 80% respectively. The expression of myc in the third trimester was mainly in syncytiotrophoblasts. The expression of myc decreased gradually and disappeared in the full-term. The expressions of ras and myc in hydatidiform mole were 52% and 60%, respectively. The expression of myc gene was related to the proliferation of trophoblast (P <001), and the expressions of ras and myc were related to the malignant transformation of hydatidiform mole (P <001, P <005). Conclusion: The overexpression of ras and myc is associated with the malignant transformation of hydatidiform mole.
