目的 了解神经干细胞分化前后的差异表达基因 ,为神经干细胞的分化调控提供基础研究资料。方法 利用细胞培养、DNA微阵列技术等 ,检测了神经干细胞分化前后的差异表达基因。结果 检测到了 10 0 0多个差异表达基因 ,其中明显上调的基因 13 8个 ,明显下调的基因 179个 ,基因种类很多如细胞周期相关基因、发育与分化相关基因、受体与通道相关基因、细胞骨架相关基因、生长因子相关基因、原癌基因和抑癌基因、转录和翻译调节基因、ESTs、代谢调节、蛋白运输相关基因等。结论 大量基因参与了神经干细胞的分化调控 ,其确切调控机制 ,还有待于进一步深入研究
Objective To know differentially expressed genes of neural stem cells before and after differentiation and to provide basic research data for the regulation of differentiation of neural stem cells. Methods The differentially expressed genes of neural stem cells before and after differentiation were detected by cell culture and DNA microarray. Results A total of more than 100 differentially expressed genes were detected. Among them, 138 genes were significantly up-regulated and 179 genes were significantly down-regulated. There were many gene types such as cell cycle related genes, developmental and differentiation related genes, receptor and channel related genes, Cytoskeleton related genes, growth factor related genes, proto-oncogenes and tumor suppressor genes, transcription and translation regulation genes, ESTs, metabolism regulation, protein transport related genes and the like. Conclusion A large number of genes involved in the differentiation of neural stem cells, the exact regulatory mechanism remains to be further studied
