目的观察大鼠尾吊模拟失重3周及解悬吊恢复8周后骨密度(BMD)及骨力学(biomechanics)的变化情况,探讨五加补骨方对其干预效果。方法雄性Wistar大鼠,按体重随机分为4组:对照组(Con)、模型组(Mod)、五加全程给药组(A),五加恢复期给药组(B)。A组全程给予五加补骨方水煎液灌胃,B组在8周恢复期给予与A组等剂量的五加补骨方水煎液灌胃。3周尾吊结束时及解悬吊8周时,取材测量不同部位BMD及骨力学性能。结果与对照组相比,尾吊3周后模型组、A组和B组的股骨、胫骨、第四腰椎BMD显著下降(P<0.001),股骨最大载荷和弹性载荷也显著下降(P<0.01);A组的BMD下降幅度明显小于模型组和B组。解悬吊8周后,和对照组相比,模型组股骨、胫骨BMD仍未恢复至正常水平(P<0.05);A组和B组大鼠的股骨,胫骨,第四腰椎BMD已恢复至正常水平(P>0.05);模型组,A组和B组股骨和肱骨最大载荷和弹性载荷同对照组比无显著变化。结论尾吊3周模拟失重可以造成大鼠承重骨BMD和骨力学显著降低,而五加补骨方具有明显对抗骨丢失的作用;在恢复期,骨密度及骨力学恢复缓慢,五加补骨方在一定程度上可以促进大鼠骨丢失的恢复。全程给予五加补骨方对抗骨丢失的疗效明显好于解悬吊后再给药。
Objective To observe the changes of bone mineral density (BMD) and biomechanics after simulated tail loss in rats for 3 weeks and suspension recovery for 8 weeks, and to investigate the effect of Wujia Buji Decoction on it. Methods Male Wistar rats were randomly divided into 4 groups according to body weight: control group, model group, five plus full course group (A), five plus recovery group (B). Group A was given Wujiaba Decoction decoction orally, Group B in the recovery period of 8 weeks and Group A was given the same dose of Wujiabu Decoction decoction. At the end of 3-week tail suspension and 8-week suspension, the BMD and bone mechanical properties of different parts were measured. Results Compared with the control group, the BMD of femur, tibia and fourth lumbar vertebra decreased significantly (P <0.001) and the maximum and elastic load of femur decreased significantly in model group, A group and B group ); The decrease of BMD in group A was significantly less than that in model group and group B. Compared with the control group, BMD of the femur and tibia of model group did not recover to normal level (P <0.05) after 8 weeks of solution suspension. BMD of femur, tibia and fourth lumbar vertebra of group A and B were restored to (P> 0.05). The maximum load and elastic load of femur and humerus in model group, A group and B group had no significant change compared with the control group. CONCLUSION: Three weeks after tail suspension, simulated weightlessness can significantly reduce BMD and bone mechanics of the bearing bone in rats, and the effect of Wujia Bu Ke Gui on significantly reducing bone loss. During recovery, bone mineral density and bone mechanics recover slowly, Side to a certain extent, can promote the recovery of rat bone loss. The whole course of treatment given Wu Jia Bu Bu Gu Fang against bone loss was significantly better than the suspension solution after administration.
