目的检测难治性白血病患者MLL基因及免疫表型,比较免疫表型、MLL基因的相关性及对预后的影响。方法利用荧光定量技术检测MLL基因,观察白血病MLL基因的表达水平,并进行特定的抗原标记。分析其临床意义。结果 48例难治性白血病中6例检测到MLL基因,发生率为12.5%,发生率较高,MLL-AF6融合基因均为M5型,MLL-AF4融合基因均为急性淋巴细胞白血病。结论 RQ-PCR监测融合基因可以有效观察疗效,MLL基因M4型,M5型白血病表达均明显升高,M3型显著降低。免疫表达异常及MLL基因重排其临床缓解率低,疗效差。
Objective To detect MLL gene and immunophenotype in patients with refractory leukemia and to compare the correlation between immunophenotype, MLL gene and prognosis. Methods The MLL gene was detected by fluorescence quantitative method and the expression of MLL gene in leukemia was observed and labeled with specific antigen. Analyze its clinical significance. Results The MLL gene was detected in 6 out of 48 cases of refractory leukemia, the incidence was 12.5%, the incidence was high, the MLL-AF6 fusion gene was M5, and the MLL-AF4 fusion gene was all acute lymphoblastic leukemia. Conclusion The results of RQ-PCR in monitoring the fusion gene can be effectively observed. The expression of MLL gene M4 and M5 leukemia were significantly increased, while M3 was significantly decreased. Immune dysplasia and MLL gene rearrangement of its clinical remission rate is low, poor efficacy.
