目的研究髓过氧化物酶(MPO)在各类型急性非淋巴细胞白血病(ANLL)中表达的差异,探讨MPO表达的定量检测在ANLL分型诊断中的意义。方法采用毛细管电泳(CE)电化学检测法检测36例ANLL患者及30例正常骨髓单个核细胞(BMMNC)中MPO表达。结果MPO在ANLL中的表达显著高于其在正常对照中的表达(P<0·05);M3病例的MPO值最高,与其他4组比较差异有显著性(P<0·05);M2、M4其次,与其他3组比较差异有显著性(P<0·05);M1再次,与其他4组比较差异有显著性(P值均<0·05);M5最低,与其他4组比较差异有显著性(P<0·05)。M6患者只有2例,未进行统计学分析,其平均值介于M4与M1之间。各亚型中MPO值,符合M2b>M2a、M4a>M4b和M5b>M5a趋势。结论毛细管电泳电化学检测作为一种新的检测方法,首次实现了对MPO的定量检测,灵敏度高,将其应用于ANLL的分型诊断是可行的。
Objective To investigate the difference of myeloperoxidase (MPO) expression in various types of acute non-lymphocytic leukemia (ANLL) and to explore the significance of the quantitative detection of MPO expression in the diagnosis of ANLL. Methods The expression of MPO in 36 patients with ANLL and 30 normal bone marrow mononuclear cells (BMMNC) was detected by capillary electrophoresis (CE) electrochemical detection. Results The expression of MPO in ANLL was significantly higher than that in normal control (P <0.05); the MPO in M3 was the highest, and the difference was significant compared with other 4 groups (P <0.05); M2 (P <0.05); M1 again, compared with the other four groups were significantly different (P <0.05); M5 lowest, compared with the other four groups The difference was significant (P <0.05). Only two patients with M6 did not perform statistical analysis, with an average between M4 and M1. The MPO values of each subtype met the trend of M2b> M2a, M4a> M4b and M5b> M5a. Conclusion As a new detection method, capillary electrophoresis electrochemical detection is the first time to achieve quantitative detection of MPO with high sensitivity. It is feasible to apply it to the diagnosis of ANLL.
