目的探讨膀胱癌中上皮钙黏附素(E-cd)和树突细胞(DC)的表达及其与生物学行为的关系。方法免疫组织化学检测采用链霉素-生物素(LSAB)免疫酶染色法,对65例膀胱癌手术切除标本中的E-cd、DC的表达进行了对照检测,染色时用已知E-cd和S100阳性的正常膀胱黏膜及膀胱肌层内神经作阳性对照,用PBS取代一抗作阴性对照。结果65例膀胱癌中33例E-cd正常表达(50.8%),20例DC正常表达(30.8%),E-cd正常表达率在Ⅰ、Ⅱ和Ⅲ级肿瘤中分别为70.1%、66.6%和25.9%,在T_(is-1)期和T_(2-4)期肿瘤中分别为80.6%和23.5%;DC正常表达率在Ⅰ、Ⅱ和Ⅲ级肿瘤中分别为76.5%、33.3%和0,在T_(is-l)期和T_(2-4)期肿瘤中分别为63.3%和2.7%。在不同病理分级和临床分期的膀胱癌中,E-cd和DC的正常表达率差异有统计学意义(P<0.05)。结论E-cd和DC的正常表达率均随膀胱癌的病理分级、临床分期的上升而下降,E-cd和DC正常表达者的肿瘤复发率和死亡率均显著低于异常表达表。E-cd和DC可作为预测膀胱癌生物学行为的指标之一。
Objective To investigate the expression of E-cadherin and dendritic cells (DCs) in bladder cancer and its relationship with the biological behavior. Methods Immunohistochemical staining was used to detect the expression of E-cd and DC in 65 cases of bladder cancer surgically removed by streptavidin-biotin (LSAB) immunoenzyme staining. The staining was performed with known E-cd And S100-positive normal bladder mucosa and bladder myenteric nerve as a positive control, with PBS instead of primary antibody as a negative control. Results The positive expression rate of E-cd in 33 cases (50.8%) of 65 cases of bladder cancer and 20 cases of normal expression of DC (30.8%), the normal expression rate of E-cd in grade Ⅰ, Ⅱ and Ⅲ tumors were 70 .1%, 66.6% and 25.9% respectively, which were 80.6% and 23.5% respectively in T-is-1 and T-2, , 76.5%, 33.3% and 0% of grade II and grade III tumors respectively, which were 63.3% and 2.7% respectively in T-1 and T 2-4 tumor stage, . In different pathological grades and clinical stages of bladder cancer, the normal expression rates of E-cd and DC were significantly different (P <0.05). Conclusions The normal expression rates of E-cd and DC both decrease with the rise of clinical stage and the pathological grade of bladder cancer. The recurrence rate and mortality of E-cd and DC were significantly lower than those of abnormal expression. E-cd and DC can be used as predictors of biological behavior of bladder cancer.
