本文介绍了应用血管灌流法,制备脊髓、脑髓和内耳等的电镜标本技术。灌流固定液为2%三聚甲醛—2.5%戊二醛混合液。灌流液经由腹主动脉流入脊髓,经由颈总动脉和椎动脉进入脑髓和内耳。经过血管灌流之后,从脊柱、头骨和颞骨等取出标本,标本经定向取材之后,再以1%O_sO_4进行后固定。欲想观察标本的某一特定切面时,可进行定向包埋。应用血管灌流法制作的脊髓、脑髓和内耳等的细胞超微结构保存得皆较完好。
This article describes the application of vascular perfusion method, preparation of spinal cord, brain and inner ear and other electron microscopy specimen technology. Perfusion fixed solution is 2% trioxane - 2.5% glutaraldehyde mixture. Perfusion fluid flows through the abdominal aorta into the spinal cord, via the common carotid artery and vertebral artery into the brain and inner ear. After vascular perfusion, specimens were removed from the spine, skull and temporal bone, and the specimens were oriented and then fixed with 1% O_sO_4. If you want to observe a particular section of the specimen, it can be targeted for embedding. Application of vascular perfusion method of spinal cord, brain and inner ear cells and other cell ultrastructure preservation are better.
