为了进一步全面地阐明荆芥药材乙醇提取物中主要发挥抗肺癌作用的化学组成,本文基于微流控芯片技术开展不同产地荆芥药材乙醇提取物的体外抗肺肿瘤药效学研究,并通过HPLC方法建立9种不同产地荆芥药材乙醇提取物的指纹图谱,将药效指标与指纹图谱的相关信息通过灰色关联度软件进行谱效关系分析,采用HPLC-Q-TOF/MS的技术手段对其进行快速分析与鉴定。结果显示,关联系数较大的色谱峰中第19、6、16、11、18号峰所代表的化合物为香叶木素、木犀草苷、木犀草素、橙皮苷、芹菜素;第10、12、20号峰可能为柚皮素-7-O-葡萄糖醛酸苷或槲皮苷、迷迭香酸甲酯或异鼠李素-3-木糖苷、5,7-二羟基-6,4-二甲氧基黄酮。该研究进一步明确了荆芥中发挥抗肺肿瘤作用的主要化学组成,为荆芥药材的质量控制及中药复杂成分的药效物质基础研究提供方法学探讨和实验依据。
In order to further fully elucidate the chemical composition of the ethanol extract of Nepeta medicinal herbs that mainly exert anti-lung cancer effects, this paper based on the microfluidic chip technology to carry out in vitro anti-lung tumor pharmacodynamic study of ethanol extract from Nepeta medicinal materials in different areas and by HPLC Methods Nine fingerprints of ethanol extracts of Nepeta medicinal materials from different habitats were established. The correlation between pharmacodynamic parameters and fingerprints was analyzed by gray relational degree software, and the relationship between them was analyzed by HPLC-Q-TOF / MS For rapid analysis and identification. The results showed that the compounds represented by the peaks No. 19, No. 6, No. 6, No. 11 and No. 18 in the chromatographic peak with larger correlation coefficient were the geraniolin, luteolin, luteolin, hesperidin and apigenin; Peaks 12 and 20 may be naringenin-7-O-glucuronide or quercitrin, methyl rosinate or isorhamnetin-3-xyloside, 5,7-dihydroxy- 4-dimethoxyflavone. The study further clarified the main chemical composition of Nepeta anti-lung cancer play a role for the quality control of Nepeta herbs and pharmacological substances of complex components of traditional Chinese medicine provide a methodological basis and experimental basis.
