目的观察Pokemon在肝细胞癌(HCC)、肝细胞癌远处转移、肝硬化患者和健康人群外周血中的表达特征及其影响因素。分析Pokemon作为HCC诊断、预测转移复发标记物的价值。方法研究对象分4组(n=40)。HCC合并乙型肝炎后肝硬化行肝移植患者、HCC远处转移合并乙型肝炎后肝硬化、乙型肝炎后肝硬化及健康人群各抽外周血2ml 1次。用实时定量逆转录-聚合酶链反应(RT-PCR)法检测Pokemon mRNA的表达。结果(1)Pokemon在HCC、肝硬化和健康人群外周血中的表达差异无统计学意义(P>0.05)。(2)HCC远处转移组Pokemon表达量、表达比例明显高于其余各组(P<0.05)。(3)HCC外周血中Pokemon表达水平与性别、年龄、HBSAG、HBV-DNA、AST、ALT、AFP无关(P>0.05)。结论HCC患者外周血中的Pokemon表达水平不高;肝硬化、健康人群组也可能存在Pokemon的表达。HCC远处转移组Pokemon表达水平明显高于HCC肝移植、肝硬化和健康人群组。监测外周血中Pokemon表达水平可能有助于诊断和预测HCC远处转移。
Objective To observe the expression of Pokemon in peripheral blood of patients with hepatocellular carcinoma (HCC), distant metastasis of hepatocellular carcinoma (HCC), cirrhosis and healthy subjects and its influencing factors. The value of Pokemon as a marker for HCC diagnosis and prediction of metastasis recurrence was analyzed. Methods The subjects were divided into 4 groups (n = 40). HCC with hepatitis B cirrhosis in patients with liver transplantation, distant metastasis of HCC with hepatitis B cirrhosis, hepatitis B cirrhosis and healthy people pumping 2ml of peripheral blood 1 times. Pokemon mRNA expression was detected by real-time quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results (1) There was no significant difference in the expression of Pokemon between HCC, cirrhosis and healthy people (P> 0.05). (2) The expression of Pokemon in distant metastasis group was significantly higher than that in other groups (P <0.05). (3) The expression of Pokemon in HCC peripheral blood was not related to sex, age, HBSAG, HBV-DNA, AST, ALT, AFP (P> 0.05). Conclusion The expression of Pokemon in peripheral blood of patients with HCC is not high. Pokemon may also be present in cirrhosis and healthy people. Pokemon expression in distant metastasis HCC group was significantly higher than that in HCC liver transplantation, cirrhosis and healthy population. Monitoring the level of Pokemon expression in peripheral blood may help to diagnose and predict distant metastasis of HCC.
