目的优化双重免疫组织化学技术,检测乳腺癌中STAT5和ER蛋白表达的相关性,并分析两者联合检测在乳腺癌治疗方案的制定及预后评估中的参考意义。方法用不同染色顺序优化双重免疫组织化学技术,并用优化的方法检测50例乳腺癌组织中STAT5和ER蛋白的共表达。结果染色顺序为STAT5→NBT/BCIP显色→ER→DAB显色者阳性对比明显,双标记效果理想;乳腺癌肿瘤细胞STAT5-ER双标记阳性率、ER单标记阳性率均高于STAT5单标记阳性率,差异有显著性(P<0.05),而双标记阳性率和ER单标记阳性率比较无显著性差异。结论应用优化的双重免疫组织化学技术检测乳腺癌组织中STAT5和ER的相关表达,有助于指导临床预测乳腺癌的内分泌治疗反应及其预后。
Objective To optimize the double immunohistochemical technique to detect the expression of STAT5 and ER in breast cancer, and to analyze the significance of the combined detection of these two in the development of breast cancer treatment plan and prognosis evaluation. Methods Double immunohistochemistry was optimized with different staining sequences and the co-expression of STAT5 and ER proteins in 50 cases of breast cancer tissues was detected by the optimized method. Results The positive staining of STAT5 → NBT / BCIP → ER → DAB was positive, and the double labeling was effective. The positive rates of STAT5-ER double staining and ER single labeling in breast cancer were higher than those in single STAT5 Positive rate, the difference was significant (P <0.05), while the positive rate of double labeling and ER single marker positive rate was no significant difference. Conclusion The application of optimized dual immunohistochemistry in detecting the expression of STAT5 and ER in breast cancer tissue is helpful to guide clinical prediction of the endocrine response and prognosis of breast cancer.
