以MS培养基为基础 ,通过改变植物激素配比和添加附加物 ,确定了适合籼稻愈伤组织的诱导、继代和再生的优化条件。用不同的抗生素与除草剂 (Bar)处理愈伤组织 ,揭示了愈伤组织与抗生素和除草剂之间的关系 ,确定筛选具有卡那霉素 (Kan)或Bar抗性标记的转化子筛选浓度。同时通过不同抗生素的组合 ,选择出抑制共培养后的籼稻愈伤组织表面残余农杆菌合适条件。并对植物激素和抗生素应用于愈伤组织的转基因系统中作用作了进一步的讨论
Based on MS medium, the optimal conditions for induction, subculture and regeneration of indica rice callus were determined by changing the ratio of plant hormones and adding additives. The callus was treated with different antibiotics and herbicides (Bar), the relationship between callus and antibiotics and herbicides was revealed, and the selection of screening transformants with kanamycin (Kan) or Bar resistance markers . At the same time, through the combination of different antibiotics, we selected the suitable conditions for inhibiting the residual Agrobacterium on the callus surface of indica rice after co-culture. The role of plant hormones and antibiotics in the transgenic system of callus was further discussed
