目的研究氧化槐定碱(oxysophoridine,OSR)对福尔马林致痛大鼠脊髓背角氨基酸含量的影响。方法实验采用Wist-ar大鼠,OSR(1 000,500,250 mg.kg-1,ip)1 h后右后足跖掌部皮下注射福尔马林(5%,100μL)致痛,30 min后取脊髓L4-6节段背角组织,采用氨基酸自动分析仪检测脊髓组织氨基酸含量。结果 OSR(1 000,500 mg.kg-1)可使福尔马林致痛大鼠脊髓L4-6节段背角组织中谷氨酸含量明显下降(P<0.05),OSR(1 000 mg.kg-1)还能明显增加γ-氨基丁酸和甘氨酸含量(P<0.05,P<0.01)。结论 OSR能降低福尔马林致痛大鼠脊髓L4-6节段谷氨酸含量并升高γ-氨基丁酸和甘氨酸含量,此作用与OSR的镇痛作用有关。
Objective To study the effect of oxysophoridine (OSR) on the content of amino acids in spinal dorsal horn of formalin-induced rats. METHODS: Wist-ar rats were used. After 1 h of OSR (1 000, 500, 250 mg.kg-1, ip), formalin (5%, 100 μL) was given by subcutaneous injection of formalin (5%, 100 μL). The spinal cord was taken 30 minutes later. L4-6 segmental dorsal horn tissue was detected by amino acid automatic analyzer. Results OSR (1, 000,500 mg.kg-1) decreased the content of glutamate in the dorsal horn of spinal cord L4-6 in formalin-induced rats (P<0.05). OSR (1 000 mg.kg- 1) The content of γ-aminobutyric acid and glycine was also significantly increased (P<0.05, P<0.01). Conclusion OSR can reduce L4--6 glutamate content and increase γ-aminobutyric acid and glycine content in rat spinal cord induced by formalin. This effect is related to the analgesic effect of OSR.
