目的探讨缬沙坦对人脐动脉平滑肌细胞PAPP-A、MMP-9表达的影响。方法分别在10-5mol/L的Ox-LDL的基础上,给予AngⅡ以及AngⅡ、缬沙坦混合培养人脐动脉平滑肌细胞,以普通培养基培养为对照,应用RT-PCR法检测PAPP-A、MMP-9的表达量。结果AngⅡ组PAPP-A与MMP-9的表达量均显著高于对照组、Ox-LDL及缬沙坦组(P<0.01),缬沙坦组PAPP-A的表达量高于对照组(P<0.01),而与Ox-LDL组无明显差异(P>0.05),缬沙坦组MMP-9的表达量与Ox-LDL组及对照组均无明显差异(P>0.05)。结论AngⅡ可促进人脐动脉平滑肌细胞PAPP-A和MMP-9的表达,缬沙坦可以抑制AngⅡ的这种作用。
Objective To investigate the effects of valsartan on the expression of PAPP-A and MMP-9 in human umbilical artery smooth muscle cells. Methods Human umbilical artery smooth muscle cells were cultured with AngⅡ, AngⅡand Valsartan on the basis of 10-5mol / L Ox-LDL, and cultured in normal medium as control. The expression of PAPP-A, The expression of MMP-9. Results The expression of PAPP-A and MMP-9 in AngⅡgroup was significantly higher than that in control group, Ox-LDL and valsartan group (P <0.01), and PAPP-A in valsartan group was higher than that in control group <0.01), but no significant difference with Ox-LDL group (P> 0.05). The expression of MMP-9 in Valsartan group was not significantly different from Ox-LDL group and control group (P> 0.05). Conclusion Ang Ⅱ can promote the expression of PAPP-A and MMP-9 in human umbilical artery smooth muscle cells. Valsartan can inhibit this effect of AngⅡ.
