目的:观察脑缺血后iNOS基因表达情况。方法:在建立SHRMCAO局灶性脑缺血模型基础上,采用半定量RT-PCR技术观察脑缺血后不同时相iNOSmRNA表达水平变化。结果:显示脑缺血后1h、4h缺血区iNOSmRNA基因无明显改变,12h时iNOSmRNA基因表达升高(P>0.05),24h、48h后iNOSmRNA基因表达明显增加(P<0.01)。结论:脑缺血后中、晚期iNOS基因表达增加,其产生的NO可能参与脑缺血后期的继发性病理改变。
Objective: To observe the expression of iNOS gene after cerebral ischemia. Methods: On the basis of established SHRMCAO model of focal cerebral ischemia, semi-quantitative RT-PCR was used to observe the changes of iNOS mRNA expression at different phases after cerebral ischemia. Results: The expression of iNOS mRNA was not significantly changed at 1h and 4h after cerebral ischemia, and the expression of iNOS mRNA increased at 12h (P> 0.05). The expression of iNOS mRNA increased significantly at 24h and 48h (P <0.01) . CONCLUSION: The expression of iNOS gene increases in middle and late stage of cerebral ischemia, and the NO production may be involved in the secondary pathological changes of cerebral ischemia.
