目的:观察去甲斑蝥素-N-乳糖酰壳聚糖/丝素蛋白微球(norcantharidin-N-chitosan-silk fibroinmicrospheres,NCTD-N-CS/SF-MS)其对兔VX2肝移植瘤的化疗栓塞术后抑瘤作用。方法:取经超声引导下于肝脏左叶植入VX2肿瘤组织块建模成功的新西兰大白兔30只并随机分为5组,股动脉穿刺插管后经肝动脉分别给予载药微球(A组)、空白微球(B组)、空白微球+NCTD溶液(C组)、NCTD溶液(D组)及生理盐水(对照),术后通过血液分析、抑瘤作用、影像学检查等方式评价治疗效果。结果:各治疗组动物术后均有不同程度白细胞数量增多及肝功能的一过性损害;A组较余各组作用更强,抑瘤率高达85.01%,具有统计学意义(P<0.05),肿瘤体积远小于其他各组,肿瘤坏死率达56.78%,高于B组的49.63%及C组52.23%。肝动脉造影示栓塞彻底,仅少量侧支血管形成。A组平均生存时间最长,达到36.25 d,生命延长率达54.25%。结论:NCTD-N-CS/SF-MS可致肿瘤坏死,抑瘤作用明显(P<0.05)。
Objective: To observe the effect of norcantharidin-N-chitosan-silk fibroin microspheres (NCTD-N-CS / SF-MS) on the chemotherapy of VX2 liver xenografts in rabbits Anti-tumor effect after embolization. Methods: Thirty New Zealand white rabbits successfully implanted with VX2 tumor tissue in the left lobe of the liver under ultrasound guidance were randomly divided into 5 groups. The femoral artery was punctured and the microspheres were administered via hepatic artery (Group A) ), Blank microspheres (group B), blank microspheres + NCTD solution (group C), NCTD solution (group D) and normal saline (control) after operation were evaluated by blood analysis, tumor inhibition and imaging examination treatment effect. Results: The number of leukocytes and the transient damage of liver function were increased in all treatment groups after operation. The effect of A group was stronger than that of other groups, with the tumor inhibition rate as high as 85.01% (P <0.05) The tumor volume was far less than other groups. The tumor necrosis rate was 56.78%, higher than 49.63% in group B and 52.23% in group C, respectively. Hepatic artery angiography showed complete embolization, only a small amount of collateral vessels. The average survival time of group A was the longest, reaching 36.25 days, and the life extension rate reached 54.25%. Conclusion: NCTD-N-CS / SF-MS can induce tumor necrosis with obvious anti-tumor effect (P <0.05).
