目的探讨2型糖尿病患者外周单核细胞mi R-21水平相关代谢影响因素。方法应用实时荧光定量PCR(quantitative PCR,q PCR)技术及酶联免疫法分别检测2013年10月-2014年2月门诊2型糖尿病患者及体检正常健康人外周单核细胞mi R-21相对水平及其体外培养上清液IL-1β与IL-6水平。结果与健康对照组相比,糖尿病组体重指数(Body Mass Index,BMI)、腰臀比、收缩压、糖化血红蛋白(Hb A1c)、甘油三酯(Triglycerie,TG)及空腹血糖(Fasting Plasma Glucose,FPG)水平显著升高(P<0.01),其他体格及代谢因素等无统计学差异(P>0.05)。与健康对照组相比,2型糖尿病患者组、超重组及Ⅰ型肥胖糖尿病组其mi R-21水平分别升高1.95倍、1.97倍及1.99倍(P<0.05);女性、男性糖尿病患者分别与健康女性、男性相比,其mi R-21水平分别增高1.89倍与2倍(P<0.05)。与对照组相比,糖尿病组外周单核细胞在体外与佛波酯(PMA)及脂多糖(LPS)共孵育后,培养上清液IL-1β与IL-6水平显著升高(P<0.05)。Hb A1c及FPG与mi R-21相对水平之间存在线性关系;影响大小为Hb A1c>FPG,为正影响。结论 mi R-21相对水平在2型糖尿病代谢控制水平方面具有重要作用。
Objective To investigate the influencing factors of mi R-21 related to peripheral blood mononuclear cells in type 2 diabetic patients. Methods The relative levels of mi R-21 in peripheral blood mononuclear cells from outpatients with type 2 diabetes mellitus and normal healthy subjects from October 2013 to February 2014 were detected by quantitative PCR (qPCR) and enzyme-linked immunosorbent assay (ELISA) And its in vitro culture supernatant IL-1β and IL-6 levels. Results Compared with healthy control group, body mass index (BMI), waist-to-hip ratio, systolic blood pressure, hemoglobin A1c, triglyceride TG and fasting plasma glucose (Glucose, FPG) (P <0.01). There was no significant difference in other physical and metabolic factors (P> 0.05). Compared with healthy controls, mi R-21 levels in patients with type 2 diabetes mellitus, overweight and type I obesity increased by 1.95 times, 1.97 times and 1.99 times (P <0.05), respectively; women and men with diabetes mellitus Compared with healthy women and men, their mi R-21 levels increased 1.89 times and 2 times (P <0.05) respectively. Compared with the control group, peripheral blood mononuclear cells in diabetic group incubated with phorbol esters (PMA) and lipopolysaccharide (LPS) in vitro, IL-1β and IL-6 levels were significantly increased (P <0.05 ). There was a linear relationship between the relative levels of Hb A1c, FPG and mi R-21; the impact size was Hb A1c> FPG, positive. Conclusion The relative level of mi R-21 plays an important role in the metabolic control of type 2 diabetes mellitus.
