目的 :探讨新生儿出生时淋巴细胞功能障碍的可能环节。方法 :给予新生儿脐血和成人外周血淋巴细胞抗CD3单克隆抗体、PMA和IM刺激后 ,应用3H TdR掺入法检测淋巴细胞的增殖反应 ;Westernblot法检测淋巴细胞PLCγ1的表达。结果 :抗CD3单克隆抗体刺激 ,脐血淋巴细胞增殖反应明显低于成人 ;PMA协同IM刺激后 ,脐血淋巴细胞的增殖反应明显高于应用抗CD3单抗刺激后的 ;脐血淋巴细胞PLCγ1的表达明显弱于成人。结论 :脐血淋巴细胞功能障碍可能与细胞活化的上游信号转导缺陷有关。
Objective: To explore the possible link of neonatal lymphocyte dysfunction at birth. Methods: The neonatal cord blood and adult peripheral blood lymphocytes anti-CD3 monoclonal antibody, PMA and IM stimulation, 3H TdR incorporation assay lymphocyte proliferation response; Western blot analysis of lymphocyte PLCγ1 expression. Results: The anti-CD3 monoclonal antibody stimulated the proliferation of cord blood lymphocytes was significantly lower than that of adults. The proliferative responses of cord blood lymphocytes were significantly higher after PMA and IM stimulation than those stimulated by anti-CD3 mAbs. The cord blood lymphocytes PLCγ1 The expression was significantly weaker than that of adults. CONCLUSION: Dysfunctional umbilical cord lymphocytes may be involved in the activation of upstream signal transduction defects.
