金黄色葡萄球菌A蛋白(SPA)是构成大多数金葡菌细胞壁的一种主要成分。SPA与免疫球蛋白有高度的亲和结合能力,主要是和IgG分子的T_c段非免疫专一的结合。正是利用这个特性,使得分析和纯化免疫球蛋白的方法更加简单、快速和经济。另外,SPA在免疫酶标记、免疫萤光标记和放射免疫分析等技术中能够代替“第二抗体”。因此,在免疫学中,就许多制备和分析目的而言,SPA的应用有着广阔的前途。为此人们建立了一些提纯SPA的方法。本文仅就本实验室所采用的方法以及所得到的实验结果介绍如下。
Staphylococcus aureus Protein A (SPA) is a major component of most of the Staphylococcus aureus cell walls. SPA and immunoglobulin have a high affinity binding capacity, mainly with the T_c non-immunoglobulin IgG molecules specific binding. It is with this feature that the method of analyzing and purifying immunoglobulins is simpler, faster and more economical. In addition, SPA can replace the “second antibody” in technologies such as immunoenzyme labeling, immunofluorescence labeling and radioimmunoassay. Therefore, in immunology, the application of SPA has a bright future for many preparation and analysis purposes. To this end, people have established some methods to purify the SPA. This article only on the methods used in this laboratory and the experimental results obtained are described below.
