淫羊藿总黄酮促进去卵巢大鼠骨组织Cbfal表达

目的观察淫羊藿总黄酮对去卵巢大鼠骨组织核心结合因子α1(Cbfal)表达的影响。方法54只雌性SD大鼠分成6组:假手术组、模型组、尼尔雌醇组、淫羊藿总黄酮低、中、高剂量组。对尼尔雌醇组和淫羊藿总黄酮低、中、高剂量组分别给予尼尔雌醇(0.1 mg·kg~(-1)·周~(-1))和淫羊藿总黄酮(40,80和160 mg·kg~(-1)·d~(-1))治疗12周后,以双能X线骨密度仪测定大鼠全身的骨密度;放射免疫法测定各组大鼠血清中骨钙素及雌二醇的浓度;处死各组大鼠,直接从头盖骨中提取总RNA,采用荧光定量PCR技术检测Cbfal mRNA的相对表达量。结果12周后,与假手术组相比,模型组大鼠的全身骨密度和血清雌二醇水平明显降低(均P<0.05)。淫羊藿总黄酮可以增加去卵巢大鼠的全身骨密度和血清骨钙素含量(均P<0.05),但不增加血清雌二醇的水平(P>0.05)。12周后,与假手术组相比,模型组大鼠骨组织中Cbfal mRNA表达明显降低(P<0.01)。淫羊藿总黄酮可以促进骨组织中Cbfal mRNA的表达。以淫羊藿总黄酮高剂量最为显著(P<0.01)。结论淫羊藿总黄酮对去卵巢大鼠骨质疏松症具有防治作用,其部分机制可能为淫羊藿总黄酮促进骨组织Cbfal表达,从而调节骨形成。 Objective To observe the effect of total flavonoids of Herba Epimedii on the expression of core binding factor α1 (Cbfal) in bone tissue of ovariectomized rats. Methods Fifty-four female SD rats were divided into 6 groups: sham operation group, model group, nilestriol group, epimedium total flavonoid low, middle and high dose groups. Nilestriol (0.1 mg·kg -1 ·week -1) and total flavonoids of epimedium were given to the Nylestriol group and Epimedium total flavonoid low-, medium-, and high-dose groups, respectively. After 12 weeks of treatment with 40, 80 and 160 mg·kg -1 ·d -1 , bone mineral density was measured by dual energy X-ray absorptiometry. Rats were determined by radioimmunoassay. The concentrations of serum osteocalcin and estradiol in the serum were sacrificed. Rats in each group were sacrificed and total RNA was directly extracted from the cranium. The relative expression of Cbfal mRNA was detected by fluorescence quantitative PCR. Results After 12 weeks, the total bone mineral density and serum estradiol levels in the model group were significantly lower than those in the sham group (all P<0.05). Epimedium total flavonoids increased total bone mineral density and serum osteocalcin content in ovariectomized rats (all P<0.05), but did not increase serum estradiol levels (P>0.05). After 12 weeks, compared with the sham group, the expression of Cbfal mRNA in the bone tissue of the model group was significantly decreased (P<0.01). Epimedium flavonoids can promote the expression of Cbfal mRNA in bone tissue. The highest dose of Epimedium flavonoids was the most significant (P<0.01). Conclusion Epimedium total flavonoids can prevent and treat osteoporosis in ovariectomized rats, and part of the mechanism may be that total flavonoids of Epimedium can promote the expression of Cbfal in bone tissue and regulate bone formation.
